Hydroxyapatite(HA)with copolymer of L-lactide andε-caprolactone[HA/P(LA/ CL)]is expected to be a potent biomaterial for orthopedic surgeries. Bone morphogenetic protein-2(BMP-2)has bone-inducing capability. In this in vivo study,we evaluated osteoconductivity(newly formed bone surrounding the implanted material) and osteoinductivity(newly formed bone in the implanted material)of HA/P(LA/CL) alone or combined with BMP-2 in cancellous bone in 7-month-old Sprague-Dawley rats. Surgical osteotomy in the sagittal orientation was performed on the proximal tibia and HA/P(LA/CL)alone or combined with BMP-2(1 or 10μg/block)was implanted to contact with the osteotomized cancellous bone surface. Four and eight weeks postsurgery, bone specimens at the operated sites were harvested and osteoconductivity and osteoinductivity were evaluated with quantitative histomorphometrical method. HA/P (LA/CL)block alone showed osteoconductivity from the week four but did not show osteoinductivity. Combination of HA/P(LA/CL) block and BMP-2 showed both osteoconductivity and osteoinductivity in a dose and time dependent manner. Because of its strong osteoconductivity and osteoinductivity,combination of HA/P(LA/CL)and BMP-2 might have potential clinical applications. |
